Abstract

Diabetes mellitus (DM), especially type 2, is a global health problem characterized by hyperglycemia. Oxidative stress is a major contributor to the complications of diabetes. One therapeutic strategy for managing type 2 DM is to inhibit the alpha-glucosidase enzyme. This study aims to investigate the antioxidant potential and alpha-glucosidase inhibitory activity of Portulaca oleracea L. extract, and to identify the phytochemical compounds responsible for these activities. This study was divided into 4 steps: (1) Extraction of purslane using 70% ethanol through UAE (2) Evaluation of antioxidant capacity (3) Alpha-Glucosidase Inhibitory Activity Test (4)  Quantitative screening was conducted to identify the presence of secondary metabolites using HRMS UHPLC-Q-Orbitrap. The research showed that the substance possesses antioxidant capacity, with an IC50 value of 40.41 µg/mL, and also has inhibitory effects against α-glucosidase, with an IC50 value of 229.41 µg/mL. The results of UHPLC-Q-Orbitrap mass spectrometry analysis of the 28 compounds found in the purslane extract identified flavonoids, polyphenols, alkaloids, fatty acids, terpenoids, and organic acids, including chromen-4-one, apigenin, nictoflorin, gallic acid, oleamide, 3,5-dioxooctanedioic acid, and 7-deoxyloganetin that contribute to alpha-glucosidase inhibition and correlated with antioxidant capacity. We conclude that purslane extract has potential as a natural antidiabetic agent.
